We have synthesized a 582,970-base pair Mycoplasma genitalium genome. This synthetic genome, named M. genitalium JCVI-1.0, contains all the genes of wild-type M. genitalium G37 except MG408, which was disrupted by an antibiotic marker to block pathogenicity and to allow for selection. To identify the genome as synthetic, we inserted "watermarks" at intergenic sites known to tolerate transposon insertions. Overlapping "cassettes" of 5 to 7 kilobases (kb), assembled from chemically synthesized oligonucleotides, were joined by in vitro recombination to produce intermediate (...) assemblies of approximately 24 kb, 72 kb ("1/8 genome"), and 144 kb ("1/4 genome"), which were all cloned as bacterial artificial chromosomes in Escherichia coli. Most of these intermediate clones were sequenced, and clones of all four 1/4 genomes with the correct sequence were identified. The complete synthetic genome was assembled by transformation-associated recombination cloning in the yeast Saccharomyces cerevisiae, then isolated and sequenced. A clone with the correct sequence was identified. The methods described here will be generally useful for constructing large DNA molecules from chemically synthesized pieces and also from combinations of natural and synthetic DNA segments. 10.1126/science.1151721. Speakers of English frequently associate location in space with valence, as in moving up and down the “social ladder.” If such an association also holds for the sagittal axis, an object “in front of” another object would be evaluated more positively than the one “behind.” Yet how people conceptualize relative locations depends on which frame of reference (FoR) they adopt—and hence on cross‐linguistically diverging preferences. What is conceptualized as “in front” in one variant of the relative FoR (e.g., translation) is (...) “behind” under another variant (reflection), and vice versa. Do such diverging conceptualizations of an object's location also lead to diverging evaluations? In two studies employing an implicit association test, we demonstrate, first, that speakers of German, Chinese, and Japanese indeed evaluate the object “in front of” another object more positively than the one “behind.” Second, and crucially, the reversal of which object is conceptualized as “in front” involves a corresponding reversal of valence, suggesting an impact of linguistically imparted FoR preferences on evaluative processes. Recently, a class of $\mathcal{P}\mathcal{T}$ -invariant scalar quantum field theories described by the non-Hermitian Lagrangian $\mathcal{L}$ = $ \frac{1}{2} $ (∂ϕ) 2 +gϕ 2 (iϕ)ε was studied. It was found that there are two regions of ε. For ε<0 the $\mathcal{P}\mathcal{T}$ -invariance of the Lagrangian is spontaneously broken, and as a consequence, all but the lowest-lying energy levels are complex. For ε≥0 the $\mathcal{P}\mathcal{T}$ -invariance of the Lagrangian is unbroken, and the entire energy spectrum is real and positive. The subtle (...) transition at ε=0 is not well understood. In this paper we initiate an investigation of this transition by carrying out a detailed numerical study of the effective potential V eff (ϕc) in zero-dimensional spacetime. Although this numerical work reveals some differences between the ε<0 and the ε>0 regimes, we cannot yet see convincing evidence of the transition at ε=0 in the structure of the effective potential for $\mathcal{P}\mathcal{T}$ -symmetric quantum field theories. The article analyzes the interdisciplinary methodology of city research within the philosophical and cultural approach. The authors argue that at present, besides sociological and economic approaches to the interpretation of the city, the cultural and philosophical examination of city is of special interest. It combines both theoretical issues and the practical aspects. The authors present the philosophical and cultural analysis of the city as well as the general concept of chronotopia. The concept of chronotope, proposed by M. M. Bakhtin and (...) his followers, is most often used as a tool for the analysis of fiction. According to the authors, the spatio-temporal diagnostics of the city also has significant theoretical and practical potential. The authors discuss various literary and philosophical texts analyzing the feautures and evolution of the “soul of the city” (written by Russians N.P. Antsiferov, I.M. Grevs, Y.M. Lotman, M.S. Kagan, I.I. Mitin as well as by foreign scholars Y. Slezkine, K. Schlogel, exploring the chronotope of Soviet cities). The article presents other modern Russian and foreign researchers who directly collaborated with the authors of this research in joint urban projects: M. Golovanivskaya (“syntax” of urban squares), “Ural matrix” (A. Ivanov, I. Lisovets, E. Trubina), chronotope of a small Israeli city (E. Rimon), Stanford chronotopes (J. Bender, D.E. Wellbery). Since the city, like any cultural phenomenon, manifests itself within a set of spatio-temporal coordinates, the chronotope can be considered as the basic tool for its philosophical and cultural interpretation. The article substantiates the practical aspects of applying chronotopia to the analysis of the city: chronotopes of the past and everyday life in cities of certain types, strategic opportunities and boundaries of the chronotopes of a given city. The authors concludes that the chro-notopia of city is a transdisciplinary field in the cultural and philosophical studies of the city. (shrink)

This paper reviews the uneven history of the relationship between Anthropology and Cognitive Science over the past 30 years, from its promising beginnings, followed by a period of disaffection, on up to the current context, which may lay the groundwork for reconsidering what Anthropology and (the rest of) Cognitive Science have to offer each other. We think that this history has important lessons to teach and has implications for contemporary efforts to restore Anthropology to its proper place within Cognitive Science. (...) This book explores different accounts of powers and abilities in early modern philosophy. It analyzes powers and abilities as a package, hopefully enabling us to better understand them both and to see similarities as well as dissimilarities. While some prominent early modern accounts of power have been studied in detail, this volume covers lesser-known thinkers and several early modern women philosophers. The volume also investigates early modern accounts of powers and abilities in a more systematic fashion than has been previously (...) done. By broadening its scope in these ways, the volume uncovers trends and tendencies in early modern thinking about powers and abilities that are easy to miss. The chapters explore how twenty-two early modern thinkers approached the following questions: What kind of entities are powers and abilities? Are they reducible to something categorical or not? This paper argues against the idea of climate change refugeehood. Drawing on political realism, it reconstructs the idea and function of refugeehood in international politics. Refugees are not the agencyless victims merely in search of rescue by states of the Global North, as the idea of climate refugeehood as a form of humanitarian refugeehood would have it. Nor are they simply a function of reparative justice, or of defending international state legitimacy. To liberal democracies, refugees are those fleeing political oppression. (...) They hold an important political function in inter-state relations in undermining rival political systems and strengthening liberal democratic regimes, both ideally and materially. The idea of climate refugeehood collides with the role refugeehood plays in international politics, the reasons for their admission, and the conceptualization of their plight and function. Dass solch ein einseitiges Beharren auf der Vernunft selbst ins Bose umschlagen kann, wurde in der Gestalt des Terrors der Franzosischen Revolution auf schmerzliche Weise anschaulich. Die Klassische Deutsche Philosophie erkennt, dass die Vernunft ihre wahrhafte (...) Kraft und Gute nur entfalten kann, wenn sie - anstatt das Böse zu verdrangen - sich mit dem Bösen konfrontiert. Das Vertrauen in die Vernunft ist also keineswegs gebrochen. Das Böse wird nicht als das vollkommen Fremde und Andere verstanden. Vielmehr entsteht der neue Anspruch an die Vernunft, das Böse mit dem Denken zu durchdringen. Aby Warburg wird in den letzten Jahren mehr denn je als Gründerfigur einer Kunstwissenschaft, die sich als Bildwissenschaft begreift, aber auch als Spiritus Rector einer anthropologischen Kulturwissenschaft verstanden. Sein Kreuzlinger Vortrag von 1923 zum Schlangenritual der Hopi gilt mittlerweile als Warburgs bekanntester Text. Ein Effekt dieser Popularität ist, dass sein Vortrag in den deutschen Kulturwissenschaften der prominenteste Text zu den Hopi geworden ist. Im vorliegenden Buch wird dieser Fakt einer kulturwissenschaftlichen Analyse unterzogen. Da diese nicht durch Einzelforschung bewältigt werden kann, (...) zeichnen die Beiträge des Bandes, aus verschiedenen Perspektiven - unter anderem der Philologie, Mythologie, Ethnozoologie oder Medienarchäologie - die wichtigsten Wissensformen und Transferwege des Schlangenrituals von Oraibi bis Kreuzlingen, von Walpi bis Hamburg nach. The paper provides a briefintroduction to the biotechnology revolutionand its impact upon biological researchrelevant to military uses. It describes thestatus of biological weapons today, and currentefforts to strengthen the Biological WeaponsConvention with a legally binding complianceprotocol. Specific modifications ofmicro-organisms that may be of military use arediscussed. Three examples of dual-use researchactivities are then used to highlight issuesand dilemmas in ethical decision making.

Our recent publication in Neuroethics re-constructed the perspectives of family caregivers of patients with disorders of consciousness on functional neurodiagnostics. Two papers criticized some of our methodological decisions and commented on some conclusions. In this commentary, we would like to further explain our methodological decisions. Despite the limitations of our findings, which we readily acknowledged, we continue to think they entail valid hypotheses that need further investigation. We conclude that some caregivers with high hopes for the recovery of their loved (...) ones with DOC will most likely not consider results of functional neuroimaging as guiding information for treatment decisions, despite efforts taken to deliver information to them. Caregivers of that type might argue that such test-results are not a reliable source of information for the judgement of whether their loved one is likely going to recover or not. We introduce the concept of epistemic beliefs to formulate this hypothesis and suggest that future qualitative studies in this area should be aware of such beliefs when investigating the effects of functional neurodiagnostics on knowledge communication and shared decision making for patients with DOC. This book re-examines the roles of causation and cognition in early modern philosophy. The standard historical narrative suggests that early modern thinkers abandoned Aristotelian models of formal causation in favor of doctrines that appealed to relations of efficient causation between material objects and cognizers. This narrative has been criticized in recent scholarship from at least two directions. Scholars have emphasized that we should not think of the Aristotelian tradition in such monolithic terms, and that many early modern thinkers did not (...) unequivocally reduce all causation to efficient causation. -/- In line with this general approach, this book features original essays written by leading experts in early modern philosophy. It is organized around five guiding questions: -/- What are the entities involved in causal processes leading to cognition? What type(s) or kind(s) of causality are at stake? Are early modern thinkers confined to efficient causation or do other types of causation play a role? What is God's role in causal processes leading to cognition? The volume is unique in that it explores both well-known and understudied historical figures, and in that it emphasizes the intimate relationship between causation and cognition to open up new perspectives on early modern philosophy of mind and metaphysics. School violence is a burning issue these days. This book provides an in-depth analysis of violence prevention programs and an assessment of their effectiveness, using data from observations, individual interviews, and focus groups, as well as published data from the schools. It is distinguished by its focus on the cultural and structural context of school violence and violence prevention efforts. Where most other researchers use quantitative measures, such as surveys, to assess the effectiveness of violence prevention programs, the authors of (...) this book use qualitative research and ethnography to study the environment where such programs take place. We have argued that Lehrer's definitions of coherence and justification have serious technical defects. As a result, the definition of justification is both too weak and too strong. We have suggested solutions for some of the problems, but others seem irremediable. We would also argue more generally that if coherence is anything like what Lehrer's theory says it is, then coherence is neither necessary nor sufficient for justification. While our current objections are directed at the ‘letter’ of Lehrer's theory, other (...) criticisms can be aimed at its very ‘spirit’. We would argue that coherence is unnecessary for justification because of the existence of ‘basic beliefs’, those about self-presenting states (‘I have a tingling sensation in my leg’) or self-evident truths (‘All men are men’). Such beliefs may be justified even though there are no other propositions in the subject's acceptance system that makes them more probable than competitors. Coherence is, moreover, insufficient for justification, because it ignores the inferential structure of the subject's acceptance system, and requires no justification of any kind for the subject's acceptance system itself. But we must develop these more fundamental objections on another occasion. (shrink)
