Almost all biomedical research to date has relied upon mean measurements from cell populations, however it is well established that what it is observed at this macroscopic level can be the result of many interactions of several different single cells. Thus, the observable macroscopic ‘average’ cannot outright be used as representative of the ‘average cell’. Rather, it is the resulting emerging behaviour of the actions and interactions of many different cells. Single‐cell RNA sequencing (scRNA‐Seq) enables the comparison of the transcriptomes (...) of individual cells. This provides high‐resolution maps of the dynamic cellular programmes allowing us to answer fundamental biological questions on their function and evolution. It also allows to address medical questions such as the role of rare cell populations contributing to disease progression and therapeutic resistance. Furthermore, it provides an understanding of context‐specific dependencies, namely the behaviour and function that a cell has in a specific context, which can be crucial to understand some complex diseases, such as diabetes, cardiovascular disease and cancer. Here, we provide an overview of scRNA‐Seq, including a comparative review of emerging technologies and computational pipelines. We discuss the current and emerging applications and focus on tumour heterogeneity a clear example of how scRNA‐Seq can provide new understanding of a complex disease. Additionally, we review the limitations and highlight the need of powerful computational pipelines and reproducible protocols for the broader acceptance of this technique in basic and clinical research. Working memory (WM) is a fundamental cognitive ability to support complex thought, but it is limited in capacity. WM training has shown the potential to make benefit for those in need. Many studies have shown the potential of transcranial direct current stimulation (tDCS) to transiently enhance WM performance by delivering low current to the brain cortex of interest via electrodes on the scalp. TDCS has also been revealed as a promising intervention to augment WM training in a few studies. However, (...) those few tDCS-paired WM training studies focused more on the effect of tDCS on WM enhancement and its transferability after training but paid less attention to the variation of cognitive performance during the training procedure. The current study attempted to explore the effect of tDCS on the variation of performance during WM training in healthy young adults. All the participant received WM training with the load-adaptive verbal N-back task for five days. During the training procedure, active/sham anodal high-definition tDCS (HD-tDCS) was used to stimulate the left dorsolateral prefrontal cortex (DLPFC). To examine the training effect, pre- and post-test were performed respectively one-day before and after the training sessions respectively. At the beginning of each training session, stable-load WM tasks were performed to examine the performance variation during training. Compared to the sham stimulation, higher learning rates of performance metrics during the training procedure were found when WM training was combined with active anodal HD-tDCS. The performance improvements (post-pre) of the active group were also found to be higher than those of sham group and were transferred to a similar untrained WM task. Further analysis revealed that the negative relation between the training improvements and baseline performance. These findings show the potential that tDCS may be leveraged as an intervention to facilitate WM training for those in need of higher WM ability. (shrink)

Purpose: The cognitive effects of total sleep deprivation on the brain remain poorly understood. Electroencephalography is a very useful tool for detecting spontaneous brain activity in the resting state. Quasi-stable electrical distributions, known as microstates, carry useful information about the dynamics of large-scale brain networks. In this study, microstate analysis was used to study changes in brain activity after 24 h of total sleep deprivation.Participants and Methods: Twenty-seven healthy volunteers were recruited and underwent EEG scans before and after 24 h (...) of TSD. Microstate analysis was applied, and six microstate classes were identified. Topographies and temporal parameters of the microstates were compared between the rested wakefulness and TSD conditions.Results: Microstate class A showed lower global explained variance, frequency of occurrence, and time coverage in TSD than RW, whereas microstate class D displayed higher GEV, frequency of occurrence, and time coverage in TSD compared to RW. Moreover, subjective sleepiness was significantly negatively correlated with the microstate parameters of class A and positively correlated with the microstate parameters of class D. Transition analysis revealed that class B exhibited a higher probability of transition than did classes D and F in TSD compared to RW.Conclusion: The observation suggests alterations of the dynamic brain-state properties of TSD in healthy young male subjects, which may serve as system-level neural underpinnings for cognitive declines in sleep-deprived subjects. ObjectivesAlthough socioeconomic status has been shown to be an important determinant of physical activity in adults, the association in children and adolescents remains less consistent based on evidence from western developed countries. The purpose of this study, therefore, is to investigate associations between SES and PA among Chinese children and adolescents.MethodsA cross-sectional study was conducted with a self-reported questionnaire in China. The multi-stage stratified cluster sampling method was used, and 2,955 children and adolescents were enrolled in this study. SES was (...) assessed by measuring parental education levels, perceived family wealth, and Family Affluence Scale II. PA was assessed by reliable and valid items from the Health Behavior School-aged Children Surveillance questionnaires. Descriptive statistics were used to report sample characteristics, including demographic characteristics, levels of PA, and SES. The Chi-square test was applied to analyze the differences of PA in gender and school period. Binary logistic regression was used to analyze the association between SES and PA among children and adolescents.ResultsOverall, 10.4% of children and adolescents accumulated moderate-to-vigorous physical activity at least 60 min per day. Boys were more physically active than girls. Higher socioeconomic status was positively associated with higher physical activity levels among children and adolescents, especially using FAS and mother education level as SES measures. The association between SES and PA varied by gender and grade group, and the effects of SES are also different on weekdays and weekends.ConclusionThis study found socioeconomic disparities in PA among Chinese children and adolescents, and showed the importance of targeting children and adolescents with low SES intervention priority. Based on these research findings, it is suggested that SES, especially for FAS and mother education level, should be considered when designing and implementing the promotion of regular PA in children and adolescents. Health policymakers may use this information to develop interventions to reduce health inequalities among children and adolescents in the future. (shrink)

In traditional reward systems, managers’ key performance indicators must surpass an agreed target in order to achieve a financial bonus. This system is designed to motivate high performance; yet adverse behavioural effects are often observed. These include middle managers becoming incentivised to game the reward system and target negotiation process they are subject to. This paper discusses three approaches to preventing such undesirable behavioural effects: Firstly, a linear pay-for-performance system without a target floor for receiving a performance bonus. Secondly, a (...) bonus banking system where managers participate at the company’s losses as well as its gains. Thirdly, a proposal that uses the Beyond Budgeting approach which radically negates the core premises of the traditional reward system and replaces rewards for individual manager performance with those for wider team performance. These three approaches are compared and evaluated with respect to their potential to prevent unwanted manager behaviour, and on consideration of how effectively they may be operationalised. (shrink)

BackgroundMild cognitive impairment is a common neurological disorder. Moxibustion has been shown to be effective in treating MCI, but its therapeutic mechanisms still remain unclear. This study mainly aimed to investigate the modulation effect of moxibustion treatment for patients with MCI by functional magnetic resonance imaging.MethodsA total of 47 patients with MCI and 30 healthy controls participated in resting-state fMRI imaging scans. Patients with MCI were randomly divided into true moxibustion group and sham moxibustion group. The degree centrality approach was (...) applied to distinguish altered brain functions. Correlation analysis was then performed to examine the relationships between the neuroimaging findings and clinical symptoms.ResultsCompared with HCs, patients with MCI mainly showed decreased DC in the left middle frontal cortex and bilateral middle cingulate cortex. After moxibustion treatment, the SHAM group had no significant DC findings, while TRUE group mainly showed significant increased DC in the bilateral MFC and MCC, as well as decreased DC in the left middle occipital cortex. Repeated measures analysis of variance showed significant interactions between the two groups of patients with MCI. In addition, the higher Mini-Mental State Examination score was significantly positively correlated with increased DC in the right MFC and left MCC after moxibustion treatment.ConclusionOur findings demonstrate that the potential value of moxibustion treatment on MCI, which adds new insights into the popular view that moxibustion treatment may slow cognitive decline in patients with MCI. This paper examines the concept of Rendaozhuyi in Late Qing and Early Republican China. Appearing as early as 1903, Rendaozhuyi is the Chinese rendering of both humanism and humanitarianism. For the Chinese intellectuals during the Late Qing and Early Republican period, “rendao” itself represented a modern value of humanity and human dignity. In the wake of the Great War, Rendaozhuyi gained tremendous popularity among the May-Fourth scholars. Some of them held it up as a universal ideal and tool to critique (...) Chinese tradition, while others respectfully disagreed, worrying it would undermine the collective morale of “strengthening the nation”. Background:As caring in times of pandemics becomes extremely stressful, the volume and intensity of nursing work witness significant increase. Ethical practices are therefore even more important for nurses and nurse leaders during this special period.Research aim:The aim was to explore the relationship between ethical nurse leaders and nurses’ task mastery and ostracism, and to examine the mediating role of relational identification in this relationship during pandemics.Research design:Based on social exchange theory, this study tests a theoretical model proposing that ethical nurse (...) leaders can increase nurses’ task mastery and reduce their ostracism by improving their relational identification with leaders during pandemics.Participants and research context:A multilevel and multi-wave field study using data from 172 nurses from 45 departments of two comprehensive hospitals was performed from April to August 2020 to test proposed hypotheses.Ethical considerations:We received formal approvals from the ethical committee of the hospital where we conducted this study before the data collection.Results:Ethical nurse leaders can indeed increase nurses’ task mastery and reduce their ostracism during the pandemic period; furthermore, nurses’ identification with their leaders mediates these relationships. We find that ethical leadership plays an even more important role in improving nurses’ task mastery and reducing their ostracism that may be facilitated by pandemics this special time. Nurses will become more identified with their leaders when they are treated by ethical ways.Discussion:The study tries to advance our understanding of the important role of ethical leadership in nurse management literature and provide useful suggestions for healthcare institutions, nurse leaders, and nurses during pandemics.Conclusion:Theoretical contributions and practical implications of our findings are discussed. Specifically, we suggest that healthcare institutions cultivate ethical nurse leaders to facilitate nurses’ relational identification, which in turn will positively influence work outcomes. (shrink)

Anxious major depressive disorder is a common subtype of major depressive disorder; however, its unique neural mechanism is not well-understood currently. Using multimodal MRI data, this study examined common and specific alterations of amygdala subregions between patients with and without anxiety. No alterations were observed in the gray matter volume or intra-region functional integration in either patient group. Compared with the controls, both patient groups showed decreased functional connectivity between the left superficial amygdala and the left putamen, and between the (...) right superficial amygdala and the bilateral anterior cingulate cortex and medial orbitofrontal cortex, while only patients with anxiety exhibited decreased activity in the bilateral laterobasal and superficial amygdala. Moreover, the decreased activity correlated negatively with the Hamilton depression scale scores in the patients with anxiety. These findings provided insights into the pathophysiologic processes of anxious major depressive disorder and may help to develop new and effective treatment programs. (shrink)

In this paper, we employ the unique qualified foreign institutional investors (QFII) scheme in China to investigate whether and how the different religious beliefs in the areas where foreign institutional shareholders from are associated with the corporate social responsibility (CSR) performance of domestic firms. After controlling for other determinants, we find robust evidence that firms with QFII investors from areas with stronger religious beliefs have better CSR performance than those that do not have these beliefs'. This association is more pronounced (...) when a QFII has a shorter holding period, has a relatively large ownership in the firm, or is a more committed investor in China. The above moderating results show that the stock preference may be a channel through which religious QFIIs affect firms’ CSR performance. Our paper contributes to the growing body of literature on CSR and on the effects of investors’ religious beliefs. It also offers useful guidance to listed companies, institutional investors, regulators, and other stakeholders. (shrink)

ObjectiveThe aim of our study was to investigate abnormal changes in brain activity in patients with persistent postural-perceptual dizziness using magnetoencephalography.MethodsMagnetoencephalography recordings from 18 PPPD patients and 18 healthy controls were analyzed to determine the source of brain activity in seven frequency ranges using accumulated source imaging.ResultsOur study showed that significant changes in the patterns of localization in the temporal-parietal junction were observed at 1–4, 4–8, and 12–30 Hz in PPPD patients compared with healthy controls, and changes in the frontal (...) cortex were found at 1–4, 80–250, and 250–500 Hz in PPPD patients compared with controls. The neuromagnetic activity in TPJ was observed increased significantly in 1–4 and 4–8 Hz, while the neuromagnetic activity in frontal cortex was found increased significantly in 1–4 Hz. In addition, the localized source strength in TPJ in 1–4 Hz was positively correlated with DHI score, while the localized source strength in frontal cortex in 1–4 Hz was positively correlated with HAMA score.ConclusionOur results demonstrated that alterations in the TPJ and frontal cortex may play a critical role in the pathophysiological mechanism of PPPD. The neuromagnetic activity in TPJ may be related to dizziness symptom of PPPD patients, while the neuromagnetic activity in frontal lobe may be related to emotional symptoms of PPPD patients. In addition, frequency-dependent changes in neuromagnetic activity, especially neuromagnetic activity in low frequency bands, were involved in the pathophysiology of PPPD. (shrink)

Despite considerable disruption of social order caused by the COVID-19 pandemic, it has also been said to contribute to positive psychological changes and influence on the perception of public life satisfaction. The present study aimed to explore the association between the COVID-19 related posttraumatic growth and life satisfaction and the mediating role of personal values. A two-wave longitudinal design was used. 226 self-quarantined Chinese college students completed post traumatic growth inventory, satisfaction with life scale, personal values questionnaire between February 2020 (...) and May 2021. Results showed that more than half of self-quarantined Chinese college students reported moderate to high levels of the COVID-19 related posttraumatic growth. A structural equation model revealed that COVID-19 related posttraumatic growth was positively associated to life satisfaction, and self-transcendence and self-enhancement values partially mediated this association. These findings shed light on whether and how pandemic-related posttraumatic growth influenced personal life satisfaction, supporting the outcome and process perspectives of posttraumatic growth as well as Schwartz’s value theory. Based on the findings, some positive psychology interventions, such as online rumination activities and mindfulness practice, were proposed to enhance self-quarantined college students’ posttraumatic growth and life satisfaction. Personality disorders are usually treated with face-to-face sessions and/or digital mental health services. Among many schools of therapies, schema therapy stands out because rather than simply targeting the symptoms of PD, it cordially targets the cause of PD and heals the early maladaptive schema, thus is exceptionally good at soothing emotional disturbances before enacting cognitive restructuring, resulting in long-term efficacy. However, according to Piaget’s genetic epistemology, the unmet needs lie in the fact that the schemata that determine the adaptive behavior (...) can only be formed in the interaction with the real world that the patient is living in and reconsolidated by the feedback from the object world upon the patient’s newly-formed behavior. Therefore, in order to reshape the patient’s schema modes to support adaptive behavior and regain emotional regulation capabilities of the healthy adult, one may have to reconstruct the object world surrounding the patient. Metaverse, the bestowed successor to the Internet with the cardinal feature of “the sense of full presence,” can become a powerful tool to reconstruct a new object world for the patient with the prescription of a psychotherapist, so as to transform the treatment techniques in schema therapy into the natural autobiographical experiences of patients in the new object world, thus gradually reshape the patient’s schema modes that can ultimately result in an adaptive, and more inclusive, interaction with the real world. This work describes the underlying theory, the mechanism, the process, and ethical considerations of such promising technology for the not-too-far future. (shrink)

Abstrak: Dalam pengantar pada karyanya Phenomenology of Perception, Merleau-Ponty praktis mengidentikkan filsafat dengan fenomenologi sebagai usaha untuk mempelajari kembali bagaimana cara melihat dunia. Dalam upaya tersebut ia mengajak pembaca, mengikuti slogan khas fenomenologi Husserl, untuk kembali ke permulaan atau bendabenda itu sendiri. Yang menarik adalah bahwa permulaan yang dianalisis oleh Merleau-Ponty justru tubuh manusia, sebuah dimensi yang cenderung dipandang rendah dalam sejarah filsafat Barat. Ia tidak sendirian dalam hal ini, mengingat dalam fenomenologinya Levinas juga menekankan sensibilitas sebagai locus etika. Menurut (...) penulis, gerakan fenomenologi menuju hal yang sensibel (the sensible) ini tidaklah mengubah hakikat filsafat sebagai usaha untuk mencari asal mula realitas. Realitas yang tersingkap dalam orientasi demikian justru menjadi lebih integral dan komprehensif daripada apa yang selama ini dikenal dalam sejarah filsafat dan sains. Meskipun demikian, orientasi pada pengalaman konkret manusia untuk menggali dasar realitas secara potensial menimbulkan masalah bagi fenomenologi itu sendiri yang selalu ingin kembali ke permulaan. Abstract: In the Preface to his work Phenomenology of Perception Merleau-Ponty virtually identifies philosophy with phenomenology as a way of relearning to see the world. For this purpose he invites the reader, following the catchphrase in Husserl’s phenomenology, to return to the beginning or the things themselves. What is interesting is that the beginning that Merleau-Ponty analyzes is the human body, which belongs to a dimension that tends to be despised in the history of Western philosophy. He is not alone in this type of investigation, as Levinas also emphasizes sensibility as the locus of ethics. The author argues that the phenomenological movement towards the sensible does not alter the nature of philosophy as an attempt to seek for the nature of reality. The reality as disclosed in this analysis can be more integral and comprehensive than what is usually presented in the history of philosophy and science. The orientation towards the concrete dimension of human life in search for the foundation of reality, however, may cause a problem for phenomenology itself insofar as it always tries to return to the beginning. How democratic progressive politics can happen and how it is happening in very different political arenas.

Organizational leaders are eager to unlock the creative potential of followers. Yet, there is growing evidence that creativity can also have a dark side within organizations. Building on research linking creativity and unethical behavior, we develop the construct of creative unethicality—behavior that is both unethical and novel. We draw on social exchange theory to develop a model that identifies both why and when creative unethicality emerges within organizations. Specifically, we investigate the exchange dynamics through which creative support provided by empowering (...) leaders facilitates creative unethicality under conditions of high performance pressure. Across two multi-wave, multi-source field studies with employee-coworker and leader-subordinate dyads and an experimental study with a novel unethicality measure in a business simulation, we find convergent support for our theoretical model. These findings have important theoretical and practical implications for fostering creativity in organizations without simultaneously facilitating creative unethicality. (shrink)

How, and how well, do people switch between exploration and exploitation to search for and accumulate resources? We study the decision processes underlying such exploration/exploitation trade‐offs using a novel card selection task that captures the common situation of searching among multiple resources (e.g., jobs) that can be exploited without depleting. With experience, participants learn to switch appropriately between exploration and exploitation and approach optimal performance. We model participants' behavior on this task with random, threshold, and sampling strategies, and find that (...) a linear decreasing threshold rule best fits participants' results. Further evidence that participants use decreasing threshold‐based strategies comes from reaction time differences between exploration and exploitation; however, participants themselves report non‐decreasing thresholds. Decreasing threshold strategies that “front‐load” exploration and switch quickly to exploitation are particularly effective in resource accumulation tasks, in contrast to optimal stopping problems like the Secretary Problem requiring longer exploration. A significant body of research has emerged in order to better understand unethical behavior at work and how gender plays a role in the process. In this study, we look to add to this literature by exploring how perpetrator gender influences reactions to distinct types of unethicality. Rather than viewing unethical behavior as a unitary construct, where all forms of lying, cheating, and stealing are the same, we integrate theories and concepts from the criminal justice and moral psychology literatures to (...) categorize certain unethical behaviors as either impulsive or premeditated. Given the agentic nature of premeditated unethical behavior, we draw from role congruity theory to predict that women will be punished more severely than men for their role incongruous actions. Impulsive unethical behavior, on the other hand, will be less likely to elicit perceptions of congruity or incongruity, leading to less of a gender effect. Results from three studies sampling both undergraduates and working adults in the United States, Singapore, and South Korea showed that participants were more likely to associate premeditated unethical behavior with a male perpetrator because it was seen as less feminine, and female perpetrators who engaged in premeditated unethical behavior received more severe punishment than male perpetrators due to the perceived role incongruity of their actions. Jahrhundert konkurrieren Hermeneutik, Dekonstruktion und Analytische Philosophie um die Deutungshoheit für literarische Kunstwerke. Der Vergleich der unterschiedlichen philosophischen Wahrheitstheorien kann dazu beitragen, einen Begriff der Poetik zu entwickeln, der in der Literatur mehr sieht als einen Unfall der Sprache. Im 20. Jahrhundert differenzieren sich unterschiedliche theoretische Ansätze heraus, die eine Antwort auf die Frage nach der Wahrheit der Literatur zu geben versuchen. Ausgehend von den Gründungsvätern der (...) Phänomenologie und der Analytischen Philosophie, Husserl und Frege, zeichnet die Studie diesen Prozess der Ausdifferenzierung nach, um am Beispiel von Heidegger, Gadamer, Derrida, Davidson, Rorty und Cavell die Spannung zwischen philosophischer und literarischer Wahrheit herauszustellen. Im Mittelpunkt der kritischen Auseinandersetzung mit den jeweiligen philosophischen Wahrheitstheorien steht ein Begriff der Poetik, der zwischen den unterschiedlichen Ansprüchen von Philosophie und Literatur zu vermitteln vermag.
